2.technique principle
Double antibody sandwich method was used in the kit. The detection principle is: the sample to be tested, R1 reagent, R2 reagent and M reagent are mixed for incubation, and the fluorescein (FITC) labeled G17 antibody 1 in R1 reagent and the alkaline phosphatase (ALP) labeled G17 antibody 2 in R2 reagent are combined with the G17 antigen molecules in the sample to form an immune complex. The immune complex is further captured by anti-FITC antibodies coupled to the surface of the magnetic particle and adsorbed to the surface of the magnetic particle. After washing, the unbound material was removed and the luminescent substrate was added, ALP catalyzed the luminescence of the substrate, and the relative luminescence intensity (RLU) was determined.
3.The clinical significance
Gastrin-17(G17)is a hormone secreted by the G cells of the gastric mucosa that stimulates gastric acid secretion and promotes the formation of pepsin. The clinical significance of Gastrin-17 includes:
(1. Assessment of Hyperacidity-Related Symptoms: Testing for Gastrin-17 helps in evaluating symptoms associated with excessive gastric acid, such as gastroesophageal reflux disease (GERD) and Zollinger-Ellison syndrome.
(2. Aiding in the Diagnosis of GERD: Levels of Gastrin-17 are typically elevated in cases of GERD, assisting in differentiating this condition from other causes of abdominal pain and chest discomfort.
(3. Monitoring Zollinger-Ellison Syndrome: With extremely high levels characterizing Zollinger-Ellison syndrome, serum Gastrin-17 measurement plays a crucial role in confirming the diagnosis and assessing disease activity.
(4. Evaluating Gastrinoma: Abnormal levels of Gastrin-17 may suggest the presence of a gastrinoma, a tumor involved in excessive gastrin production, usually linked to Zollinger-Ellison syndrome.
(5. Medication Management Monitoring: Under the influence of anti-secretory medications designed to reduce gastric acidity (like Proton Pump Inhibitors), Gastrin-17 levels should decrease accordingly. If they remain high despite such medication, it might indicate the possibility of a tumor.
(6. Gastric Cancer Risk Assessment: Some studies have shown that Gastrin-17 levels may increase in some patients with gastric cancer, though its specificity and sensitivity as a gastric cancer biomarker require further research.
(7. Long-Term Health Monitoring: For individuals with a family history of gastric cancer or known gastric pathologies, periodic monitoring of Gastrin-17 serves as a means of long-term health surveillance.
(8. Research into Gastrointestinal Disorders: Measuring Gastrin-17 also contributes to scientific research endeavors, elucidating the pathophysiological mechanisms behind various gastrointestinal disorders.
In summary, the assessment of Gastrin-17 is a vital step in evaluating gastrointestinal function, particularly pertaining to situations involving excessive gastric acid and potential neoplastic processes. However, the interpretation of Gastrin-17 test results requires integration with patient clinical presentations, endoscopic findings, and other laboratory work-ups to arrive at accurate diagnoses and therapeutic decisions.
4. Product features and indicators
High sensitivity: the detection limit is not higher than1.0 pmol/L;
